State the hypothesis so it's falsifiable — what exactly would count as evidence for or against it?
Specify the exact measurement and units that will be used as the primary endpoint.
Choose the statistical type; this affects sample-size thinking.
A numeric estimate used to plan effect size and sample needs. Leave blank if unknown.
What smallest change would change your decision or next step? Express as absolute units or percent.
Typical values: 0.05. Use lower alpha only when false positives are costly.
Probability of detecting the specified effect if it exists. Common choices: 0.8 or 0.9.
Provide the statistic and units used to describe spread — e.g., SD = 0.4 µmol/min.
If you've already estimated sample size, record it here and note how it was calculated.
How will you assign treatments or conditions? Describe strata or blocking if used.
Who is blinded and how? If blinding isn't possible, note why and how bias will be reduced.
Describe negative, positive, or baseline controls and why they're appropriate.
Specify the primary statistical test, transformations, and handling of outliers/missing data.
Describe exactly what outcome will be considered a success and what the next step will be.
Record any interim checks and what would stop or change the study.
Who, what, and when counts for enrollment or sample inclusion.
What will lead to exclusion and how exclusions will be recorded.
Pre-registration reduces selective reporting and increases trust.
